We are looking for a highly motivated diploma student for Leukemia research on the topic:
Collaborating events in TEL-AML1 positive acute lymphoblastic leukemia.

This project is part of the research aiming at the identification of molecular mechansims in leukemia development in the context of projects supported by FWF and GENAU-CHILD.

The research in our laboratory focuses on the etiology and molecular pathogenesis of childhood acute lymphoblastic leukemia (ALL) aimed to improve our understanding of leukemia development. Our current major emphasis is on two aspects of ALL carrying the t(12.21) chromosomal translocation: The function of the TEL-AML1 fusion gene (also known as ETV6-RUNX1) and the role of potential secondary events in leukemia development. These studies are carried out in model systems by loss and gain of function studies and are validated in primary leukemic cells as well as in animal models. The results should lead to the elucidation of mechanisms by which the fusion genes/mutations exert their specific function and the information whether the particular fusion gene or its downstream effectors can be specifically targeted.

Requirements: highly motivated master student in molecular biology/genetic,laboratory experience in molecular biology advantageous, excellent team player with the ability to work independently.

Title: Characterization of protective pneumococcal antigens for their role in the bacterial life cycle and pathogenesis

Background: Intercell AG is a successful biotechnology company focusing on vaccine development against numerous pathogens using platform technologies for genomic scale antigen identification and novel adjuvants. Streptococcus pneumoniae (Pneumococcus) is a widespread human pathogen that causes a variety of infections, many of those are life-threatening diseases such as pneumonia, bacteremia and meningitis. Despite the availability of licensed vaccines and antibiotic treatments, the morbidity and mortality attributed to S. pneumoniae remain significant in both developed and developing countries. At Intercell Pneumococcus is a target for the development of a protein-based vaccine and a humanized monoclonal antibody therapy for the treatment and the prevention of life threatening infections. Antigen identification and in vitro validation have been completed, and protective antigens are currently being prepared for human clinical trials. A research project that is ongoing already (PhD thesis) aims the better understanding of the role of these proteins in pneumococcal pathogenesis and host-parasite interaction. Based on gene deletion studies the two lead vaccine candidates have important non-redundant functions in bacterial multiplication showing reduced growth in vitro, abnormal septum formation and atypical cell shapes and lack of virulence in vivo.

Aim: Characterization of two pneumococcal antigens that are selected for vaccine development regarding their function in the bacterial cell cycle and their role in pathogenesis.
Methods:
Protein-protein interaction studies: Two Hybrid System, biochemical assays with recombinant proteins and cultures cell lines, binding studies, generation of knock-out strains
Cell cycle studies: localization using immunofluorescence and ELMI
Pathogenesis: in vivo studies of virulence and disease

Improvement of cellular productivity in recombinant CHO cells using flow cytometry and cell sorting


Objective:
• to find new strategies to use the cell sorter to quickly and efficiently isolate the highest producing subclones from a pool of transfectants
• to evaluate existing and new sorting methods

Currently existing methods to sort for high producers:
Cold Capture Surface Staining:
• a simple method, applicable to most products, however, does not always give a good correlation between the specific productivity and the fluorescent signal
Affinity matrix Surface staining:
• Reliable, but complicated (3 consecutive steps required to fix matrix onto cell surface)

New, to be developed methods:
New Affinity Matrix method
• Reduction of matrix fixation to a single step
Electroporation of cells using product specific labelled antibodies
• Electroporation allows to introduce not only plasmids, but also proteins into cells. Labelled product specific antibodies are retained only in cells that contain product
Amount of Endoplasmic Reticulum in the cell
• Plasma cells have been shown to have a significantly increased volume of ER, a similar increase in ER volume was sometimes observed in high-producer cells
Overall Protein Secretion rate
• A protocol exists that measures vesicle transport from the Golgi to the cytoplasma membrane thus giving an estimate of total secreted protein

Establishment and evaluation of cell lines with optimised
growth behaviour by directed evolution and cell sorting


The economic output of a biotechnological production processes is determined by the growth properties of the used cell line, the final achieved cell density as well as by its specific production rate. Mammalian cells, compared to recombinant bacteria and yeast, are notoriously slow and grow only to limited cell densities. We have recently established a directed evolution protocol assisted by flow-cytometric cell sorting that enabled the improvement of the final cell density and viability of a CHO host cell line, while at the same time reducing the glutamine dependency of the cells. Currently, the new host cell line is able to grow to higher final cell densities with extended viability compared to the parent cell line, but in the complete absence of glutamine in the culture medium. Glutamine is the main energy substrate of immortalised mammalian cells in culture, however, it is the source of ammonium, which causes a decrease in the sialidation of protein glycosylation. As the main reason for using mammalian cells is the fact that some proteins require the species-specific glycosylation patterns for biological activity, long in-vitro half life and low immunogenicity and as most of these parameters are dependend on terminal sialidation of the glycosylation chains, it is especially important to avoid high levels of glutamine in a production process.

The goal of the present master´s thesis will be to establish a production cell line growing in glutamine-free medium using the already established directed-evolution-protocol, characterisation of growth and viability of the resulting cell lines and evaluation of changes in productivity and in product quality (glycosylation)

Hi!

Würd ca. ab Jänner eine Diplomarbeit suchen.
Bevorzugt in den Bereichen Mobi, Virologie, QM,.... bei einer Firma zumindest mit angemessenem "Taschengeld" wäre optimal....

Leider tut sich hier schon lang nix mehr.

Hat irgendjemand Infos?
