Hallo,

kann sich jemand so ungefähr daran erinnern, was die Kasper so prüft?
Anscheinend kommen bei Ihr Theorie und bei Jungbauer großteils Rechnungen, ist das richtig?

Nachdem der ganze Beitrag gerade gelöscht wurde hier nochmal die Fragen von heute in Kurzfassung:

Kasper 20,5 Punkte
Jungbauer 18 Punkt (je Rechenbeispiel 9 Pkt)

KASPER

Worauf muss man bei einem Upstream process achten? (3 P)
- ich glaube das spielt auf die 3 Punkte cell culture design, media design und Reaktor design an?

-Vaccine production welche Punkt bei upstream wichtig? (6 P)
-Welche 2 Schritte gibt es bei vaccine production und welche Zellen werden verwendet dafür? (3P) die beiden Fragen haben sich irgendwie fast überschnitten, keine Ahnung...

-Welche Möglichkeiten gibt es einen Prozess zu betrieben? (1P)

-Nennen Sie Single use devices (3P)

-Vorteile von Disposibles nennen (3P)

Ein Beispiel mit E.coli Kultur und man soll Viskosität berechnen. Gegeben war die dry mass mit 15 g/L und Scherrate mit 10 s^-1. Frage wie die Viskosität ist und wie sie nach dem Homogenisierungsschritt ist. Letzte Unterfrage war was die Scherrate eigentlich aussagt (bzw ob sie aussagekräftig ist).

Ein Beispiel mit Protein mit 20000 g/mol präcipitiert, am Anfang c=20 mg/ml nach 1 Minute nur noch 0,2 mg/ml. Berechnen sollte man die Größe des Proteins (über Diffusionskoeffizienten und rh) und schätzen, wie viele Proteine sich zusammenlagern.


Es ist irgendwie weiterhin unklar welche Unterlagen man verwenden darf, viele waren mit Doran und Folien von Jungbauer da aber so richtig offen hat sie auch niemand gehabt. Prüfungsaufsicht ist auf jeden Fall sehr tolerant ;)

You must process E.coli cells, therefore the viscosity is relevant.
1) Calculate the viscosity of an E.coli broth with a dry cell weight of 25 gL-1 at a shear rate of 10 and 20 s-1
2) How much increases the viscosity when the cells are homogenized. Describe and justify your assumptions, which you have made.
You will precipitate an antibody from culture supernatant with 2M ammonium sulfate. The solubility oft he antibody in the culture supernatant is 100 mg*mL-1. Your salting out constant is 0,05.
1) Calculate the residual antibody concentration in your supernatant and the yield of your purification by this precipitation.
2)Repeat the calculation without neglecting the salt concentration in the supernatant. Justify your assumptions.

Da ja am Donnerstag die nächste Prfg ist:

*) C0=20mg/mL mit 20000 Da sinds 10^(-6) mol/mL
Ct=0,2mg/mL mit 20000 Da sinds 10^(-8) mol/mL
t=60sec
D im Medium is 1/10 von dem in Wasser.

*) Damit über perikinetische Formel kappa=1,65E+6 mL/(mol*s)
Perikinetische weils offenbar einfach reingeleert wird und kein Reaktor oder hochkomplexer Schas gefragt is...

*) Diffusionskoeffizient aus Tyn/Gusek
D(in Wasser)=9,95E-7 cm2/s und damit Ds=9,95E-8 cm2/s im Medium.

*) kappa = 8pi*D*Nav*dp den Partikeldurchmesser...
dp=1,1E-12 cm (das kann schon mal nicht sein, ich find aber keinen Fehler)

*) rh über Stokes-Einstein für Wasser (µ=0,001, und den D(in Wasser) bei 293K)
rh=2,16 nm (klingt realistisch)

*) Abschätzung mit Fraktal Df=2,4 zb und in (r/rh)^Df einsetzen - das geht natürlich nicht wenn der Partikelradius unfassbar kleiner ist als der Proteinradius...

Vlt gibts ja noch wen der sich damit herumplagt und schon gscheiter is als ich. Ist vlt doch orthokinetisch mit diversen Annahmen die Methode der Wahl?

17.)PEG Fraktionierung:
Gegeben zwei Geraden mit den Werten der Löslichkeit des Proteins und der Verunreinigung bei bestimmten % an PEG. (Graphik Y Achse löslichkeit von Protein und Verunreinigung vs X Achse % PEG) --> Die Werte der Geraden waren auch als Graphik gegeben.
Gegeben waren weiters die Konzentration des Proteins vor der fällung(10mg/mL) und der gewollte Yield(90%).
a) berechne die benötigte PEG Konzentration um auf den Yield zu kommen.
b) berechne die Reinheit, wenn die Konzentration der Verunreinigung gleich der Konzentration des Proteins ist.
c)Ist die Methode sinnvoll, wenn die Proteinkonzentration 1mg/ml und die Verunreinigungskonz 10mg/mL ist?

18.)Virusinaktivierung
Ich zitiere wort Wörtlich:
For Virus Inactivation you have to develop a sampling plan. You want to avoid false negative results with probability of 0,001. Your Virus titer after inactivation is -2
a)calculate the sampling volumen to meet the probability
b)discuss why this inactivation step cannot be validated and another strategie must be developed

You do a flocculation with PEG in a cstr. Shear rate war gegeben. Use the Camp-number! Upscale: You want to process 5 m³ and switch into a tubular flow reactor. Calculate the dimensions of the reactor!

Vor allem Altfragen, ein paar neue Sachen waren dabei:
zB:
two possibilites to isolate primary cells
definition of biosimilar, biobetter and bioidentical
advantages of personalized medicine (at least 3)
1000 L single use reactor, pump into liquid through an opening of 19 mm, how long does it take?

Jungbauer (kann mich nicht mehr auf den genauen Wortlaut erinnern):
1)
safety factor of a diploid cell is assumed; if one makes the same cell but haploid, does one need to remove less DNA?
2)
3 concentrations of Ammoniasulfate and its corresponding solubilty concentrations
One had to plot the log solubility against NH4SO4 molarity and against the ionic strength,
determine the salting out constant and calculate the needed NH4SO4 concentration for a yield of 90% (starting conc = 15 mg/mL protein)
3)
IEF
isomers of two different proteins of different MW (15 kDa and 200 kDa) are separated by IEF separetely; how much does the resolution differ between the two proteins
