Tasks
· “fish” genes from exotic lactic acid bacteria like Oenococcus oeni and Oenococcus kitaharae by PCR and gene walking
· Bioinformatics: get the sequences of your genes, do orthology and homology research, cladistic interpretation, identify conserved domains... publish the sequences in a database, study PTS-system
· Expression in lactic acid bacteria (self clones) with fusion tags
· Enzyme purification and characterization, Michaelis Menten Kinetics, test transglycosylase activities
· Immobilization on non-toxic polymers
· If you want, directed evolution by site directed or random mutagenesis to improve pH stability
· Hydrolyze aroma precursors from wine or fruit juices
· Sensory evaluation, aroma profiling by GC-MS, chemical nose....
Duration:
Bachelor thesis: 2 months
Master/Diploma thesis: 6 months
Requirements:
Fundamental knowledge in molecular biology and biochemistry, lab coat
