Enhancing protein production in the yeast
P. pastoris
Pichia pastoris is THE yeast for recombinant protein production. However, further strain improvements are necessary to make this system even more competitive. The approach used in this study is the systematic knock-out of genes, predicted by a genome-scale metabolic model that was developed in a cooperation with Prof. Sang Yup Lee from the Korea Advanced Institute of Science and Technology.
The start of the project will be at the beginning of 2011.
The work will include
Cloning of vectors in E. coli
Generation and screening of P. pastoris knock-out strains
Product analysis and quantification via SDS-PAGE, Western Blot or ELISA
Characterisation of the engineered strains (productivity, enzymatic assays, fermentation,...)
